Oxidative metabolism by cytochrome p450 enzymes is a primary method of drug metabolism . This system is a localized in the endoplasmic reticulum of liver and other tissues. For optimal oxidation, the cytochrome P450 monooxygenase system requires NADPH and molecular oxygen (O2). This system has broad substrate versatility, and as such is known as a universal oxidase, since it can affect virtually all lipophilic xenobiotics.
Broad substrate versatility is the result of having many different P450 isozymes, each with a broad and novel substrate specificity. Of the P450 isozymes, Cytochrome P450 3A4 is the most important in drug metabolism, and is involved in drug-drug and drug-food interactions.
